Well, it's not THAT exotic, just Hawaii (though partly filmed in Puerto Rico), and the characters are honeymooners, not backpackers. However...
Writer/Director Twohy (Pitch Black) is a competent guy.
Star Steve Zahn is funny.
Timmy "the elephant" Olyphant is creepy, especially since he has a decent store of rage (from his failed attempt at being a movie star after his stint as the teeth-gritting lawman in Deadwood) from which to give the villain a little realism (if he is the villain).
Speaking of which, the trailer doesn't give away much of the plot, which is commendable assuming what's left to discover is compelling.
The commenters on IMDB have some guesses:
1) Milla Jovovich (Sp) and Steve Zahn's character turn out to be the murderers
2) There is two killers, both in different relationships who secretly know each other
3) None of the characters are actually the killers and are only trying to kill one another through fear
